The JAK-STAT pathway is a primary intracellular signaling mechanism that mediates the effects of more than 50 cytokines and growth factors. Inhibitors of this pathway, known as or "jakinibs," are small molecules that bind to intracellular Janus kinase enzymes (JAK1, JAK2, JAK3, and TYK2), blocking the downstream activation of STAT proteins and subsequent gene transcription involved in inflammatory and immune responses.
